It can help you find a skyhold by shift-clicking. It is also used to open an ender portal by shift-clicking whilst your cursor is over an ender portal block.
Crafting
You must put an ender pearl with blaze powder to craft the Eye of Ender.

When you use the eye of ender (NOT by shift-clicking the end portal frame), you will see the eye of ender float away from you. This is because it is locating a skyhold.
